I have a 27 inch i7 iMac, and a 32 inch Samsung external monitor. I was wondering if I could run eyefinity across the two, or if I need to have at least three monitors to make it work.

Eyefinity will work on 2-6 monitors. But seeing as to how your main screen is 27 inch while your external screen is 32 inch, it's gonna look awkward when you get it to run. Also, both monitors will need to have a common resolution or two before it will work.
Also, you won't need eyefinity for a dual monitor setup. Just set one as the main display and extend it to the other.

You could run the setup but the different resolutions that will run will make everything weird and bad-looking . You should get a 27 inch cinema display to get the best eyefinity setup with a 27 inch iMac.
