On an iOS device you can register a @me.com email by going to Settings, Mail, Add Account then pick iCloud, then tap on the "Get a Free Apple ID" button, enter your location and birthday, tap next then tap "Get a free @me.com email address" and follow the screen's directions.
In Mac OS X Lion you go to System Preferences, iCloud, click the "Create an Apple ID" link, enter your location and birthday, tap next then click "Get a free @me.com email address" and follow the screen's directions.
