Is there any way to Jailbreak iPhone 3GS with iOS 4.0.1?

No, not currently at this time. Comex is working on a Jailbreak, it works, but its currently only at the command line Jailbreak state, so it would be hard for the average user. Expect it to be released in a few weeks...most likely the day the iPhone 4 white version is released.
